Facility Details

The Imaging Core (IC), formerly the Biological Electron Microscope Facility (BEMF) core, provides both internal and external users with access to state-of-the-art instrumentation, training, flow cytometry services, and advanced microscopy (light, epifluorescence, confocal and laser tweezers/microdissection microscopy, and scanning and transmission electron microscopy) on a recharge basis. Clients may be trained to use instruments independently, elect to have a technician operate the instruments in their presence, or may contract the technician to conduct the work and provide finished products. The IC also provides workshops, demonstrations, and laboratory sessions for UH undergraduate and graduate courses, staff, and faculty.

An award is made to the University of Hawaiʻi at Mānoa to enable the purchase and commissioning of a new Transmission Electron Microscope equipped with an energy dispersive X-ray spectroscopy system to be housed in the Biological Electron Microscopy Facility located on the main campus of the University. This advanced instrument will significantly enhance research and training capabilities across biology, materials science, and engineering disciplines while providing crucial educational opportunities for students annually through graduate research, undergraduate honors projects, and comprehensive training programs. The facility engages in numerous educational and outreach activities, utilizing the microscope for demonstrations in kindergarten through grade 12 science classes at local public and private schools, laboratory sessions for undergraduate and graduate courses, and specialized workshops for community college faculty and students. As the only fully equipped electron microscopy facility serving the state of Hawaiʻi, this instrument will strengthen existing partnerships across the University of Hawaiʻi system, government organizations, and industry collaborators, while supporting workforce development in advanced microscopy techniques essential for the research infrastructure and economic development of Hawai’i. The enhanced capabilities of the new TEM will advance frontier-pushing research programs across diverse scientific domains. Research activities enabled by the instrument's exceptional magnification, 0.20 nanometer resolution, and simultaneous elemental analysis capabilities include investigating physiological mechanisms enabling larval zooplankton survival in extreme sub-arctic environments, characterizing marine species dispersal and connectivity across vast Pacific oceanic distances, documenting fine-scale morphological adaptations in cave-dwelling arthropods, illuminating cellular characteristics of abundant marine bacteria, and understanding how giant viruses thrive in nutrient-poor ocean conditions. The system's integrated analytical platform will provide transformative insights into host-microbe interactions, protein topology, cellular ultrastructure, and viral-host dynamics, fundamentally advancing scientific knowledge across marine biology, evolutionary ecology, and molecular sciences. This award reflects NSF's statutory mission and has been deemed worthy of support through evaluation using the Foundation's intellectual merit and broader impacts review criteria.

SUMMARY/ABSTRACT To enable Precision Nutrition, a Consortium of Research Advancement Facilities & Training (CRAFT) is proposed that will establish new research facilities and enhance existing ones, ensuring that state-of-the- art equipment and other essential resources are available to COBRE-funded researchers to facilitate the conduct of high-quality, high-impact research employing cutting-edge technologies. New, emerging, and established Cores comprising CRAFT that include Epigenomics, Metabolomics, Lipidomics, Microbiomes, Nutrition, Metabolic Phenotyping, Microscopy/Imaging, and Bioinformatics will be integrated under a unified administrative structure, with directors of each Core forming an Executive Committee that facilitates external evaluations and prioritizes budgets annually. This novel configuration of Cores will allow greater resource flexibility and improved efficiency of research. Herein, we propose to establish a new Statistical Analysis, Nutrition & Diet Core, and to expand Epigenomics, Metabolomics, Lipidomics, Metabolic Phenotyping, Microscopy/Imaging, and Bioinformatics Core Facilities. Education, training, access to equipment and consultation are or will be readily available to investigators at the University in each of these areas, through the expertise of highly accomplished faculty and their support staff, postdocs and junior faculty colleagues. The Executive Committee will contract a third-party expert to measure and evaluate the effectiveness of each of the CRAFT Cores annually to ensure relevance and value for UH investigators. The goal of the CRAFT is to provide resources, expertise, education and training for the advancement of meritorious biomedical research provided by the CRAFT will produce the next generation of research leaders and mentors in the field of Precision Nutrition to ultimately benefit the people of Hawaii and the Pacific region through improvement of health outcomes and quality of life. To achieve this goal, we aim to provide access to the latest technologies/equipment (Aim 1), education and training (Aim 2), and consultation-based expertise (Aim 3) altogether supporting research in Epigenomics, Metabolomics, Lipidomics, Metabolic Phenotyping, Metagenomics (Microbiomes), Microscopy/Imaging, and Bioinformatics. Collectively, the availability of the CRAFT resources and expertise will substantively expand the direction and depth of research conducted by researchers and collaborating groups, and increase the quality of their research, thus positioning them to successfully attain high-impact, peer-reviewed publications and external grant support. By working with our Community Engagement and Outreach Core, we anticipate that the research activities in the field of Precision Nutrition will not only be relevant to community needs (e.g., health disparities that are related to deficiencies in nutrition) but also provide a concerted mechanism to encourage community support and education in the area of basic science, clinical, and translational research. Thus, the CRAFT is unique to Hawaii and is an essential foundation for Precision Nutrition with far-reaching implications to health research nationally.

Project Summary – CORE B: Microscopy Imaging Center for Research through Observation (MICRO) The Microscopy Imaging Center for Research through Observation (MICRO) Core of the Integrative Center for Environmental Microbiomes and Human Health (ICEMHH) will provide comprehensive imaging instrumentation and technical expertise at the University of Hawaiʻi at Mānoa (UHM) to support research that characterizes microorganisms and their relationship to each other, to host tissues or to their environment. The MICRO will incorporate and enhance the existing Biological Electron Microscope Facility that houses the only transmission electron microscope in the state, and the only scanning electron microscope at UHM supporting biological and biomedical research. The MICRO Core will provide the following: instrumentation, training and services for scanning and transmission electron microscopy, epifluorescent and confocal microscopy and laser microdissection and optical tweezers. A major goal of the COBRE support will be to modernize equipment and introduce users to new techniques, particularly rapidly changing fluorescent-imaging technologies, to assist in studies of microorganisms and their microbiomes. Relocation of the MICRO Core to a new Life Sciences Building, scheduled for completion in 2019, will provide a larger, custom-designed space for enhancing its research and training activities. The MICRO Core will be available to researchers across the UHM campus, to other local research and educational institutions, and to visiting scientists. The MICRO Core will favorably impact COBRE investigators and their affiliates by providing essential infrastructure and expertise to advance their research and foster collaborations that will positively impact the careers of junior investigators and more established investigators. The MICRO Core will support the growth of the ICEMHH, and (i) bring it visibility through its research and training efforts, (ii) staying abreast of advancing instrumentation and technologies in microscopy, and (iii) facilitation of outreach activities to characterize Hawaii's microbiomes.
