The WVU Flow Cytometry & Single Cell Core Facility (FCSCCF) provides instrumentation, training and scientific expertise for single cell analysis and sorting. It is equipped with three cytometers: a Becton-Dickenson (BD) FACSAria III high-speed bench top sorter, a BD LSRFortessa with 4 lasers, a high throughput sampling module, and a detector to analyze submicron-sized particles and a Guava easyCyte HT. For single cell separation, the facility has a Fluidigm C1 Single Cell Auto Prep System which isolates RNA/DNA/microRNA and generates cDNA (if needed) for downstream RNAseq, DNAseq or PCR from single cells. Other instrumentation in the WVU FSCSSF include a Miltenyi Biotec gentleMACS Otto Dissociator for disassociation of tissues into single cells or homogenization for molecular biology applications, a Miltenyi Biotec AutoMACS located in a tissue culture hood for sterile magnetic cell sorting and a Meso Scale Discovery QuickPlex SQ120 automated plate reader that detects multiple analytes by electrochemilumiscence. For nanoparticle analysis, the facility has a Malvern Panalytical NanoSight NS300 and a Zetasizer NanoZ. The facility has auxiliary laboratory space with appropriate equipment to train personnel and support the needs of researchers. The WVU FCSCCF has an analysis workstation as well as a site license for the the flow cytometry analysis software FCS Express (De Novo Software).
The facility is managed by the Director Kathleen Brundage, Ph.D who has training from the Bowdoin Flow Cytometry Course, from BD Biosciences, from Fluidigm and >30 years of experience with flow cytometry. She oversees the day-to-day operations of the facility, operates the FACSAria for cell sorting, provides necessary instrument and software training, and consults on experimental design and data analysis. The facility is house in an approximately 850 sq ft lab space located on the 2nd floor of the Robert C. Byrd Health Sciences Center North building.
